The chief physiologic property is that of relaxing smooth muscle of all kinds, including the coronary arteries, without interfering with the normal contraction. The drug has a slight analgesic effect; in larger doses, there may be a sedative effect as well. The drug is well absorbed in the gastrointestinal tract and is probably completely destroyed in the body. Its toxicity is low and there are few contraindications to its use. Less serious effects include nausea, vomiting, pallor, and sweating. Intraocular pressure may be increased, contraindicating the use of nitrites in the presence of glaucoma.

Continued use of any one of the members of the nitrite group may lead to tolerance which begins in a few days and is fully developed in two to three weeks. Nitrites have recently been suggested as a means of producing a provocative test in patients with migraine, in that their administration may precipitate a headache.
